Season ticket holders risk their season ticket being blocked for renewal if they sell them to away supporters.
They are also reminding fans it is illegal to move tickets on for financial profit or knowingly exchange them for personal gain.
A spokesman for the club said: "Wigan Athletic Football Club and the police are aware of the potential for tickets to be sold on for the sold-out Manchester United game.
"Over the past two months the club has been working with the police to do all they can to prevent this from happening.
"Firstly, tickets for the game have only been sold to season ticket holders and fans who have attended at least four home games this season.
"All tickets have the name, address and details of the purchaser on them and are being delivered to registered addresses only via secure post.
"Wigan Athletic would like to take this opportunity to remind its supporters that ticket touting is illegal and anyone caught exchanging tickets risk further proceedings."
